A. Yani Km 36 Banjarbaru, Kalimantan Selatan Email : arnida01@unlam.co.id Abstract Malaria is one of the health problems that can cause death. Manuran (Coptosapelta tomentosa Valeton ex K.Heyne) is a plant from Kotabaru South Borneo used traditionally as anti malaria. This study aims to determine the chemical content and inhibitory activity of heme polymerization based on IC50 ethanol extract of C. tomentosa’s stem. Based on the identification of chemical content with phytochemical screening and Thin Layer Chromatography shows the presence of flavonoids, terpenoids, saponins, tannins and anthraquinones in C. tomentosa’s stem. Inhibition activity of hemolymerization polymerization as early screening of antimalarial activity showed IC50 ethanol extract of C. tomentosa at 1.56 ± 0.07 mg/mL smaller than chloroquine diphosphate 1.82 ± 0.10 mg / mL. This shows that the ethanol extract of C. tomentosa’s stem has inhibition activity of hem polymerization. The result of independent test of t-test sample obtained significance value of 0.395 so that H0 is accepted, there is no significant difference (p> 0,05) between IC50 ethanol extract of C. tomentosa’s stem with positive control. Therefore, the ethanol extract of C. tomentosa’s stem can be categorized as having equivalent activity to positive control inhibiting hem polymerization.
